Studying gene expression encodes surface antigens of pathogenic flagella in Vietnam’s buffaloes and cows

In Vietnam, Trypanosomiasis has been appeared in many areas of the whole country by Trypanosoma evansi (T.evansi) with the incidence of buffaloes around 13-30%, cows around 7-14%, mortality around 6.3 - 20%.

Many studies showed RoTAT1.2 antigen presented in most of the VAT (Variable Antigen Type) of T.evansi. The scientists from Hanoi Open University, Hanoi Polytechnic University and Thai Nguyen University of Agriculture and Forestry determined that this antigen can be used to generate antibodies or apply directly to the development of the T.evansi accurate diagnosis method.
In this study, the gene encoding the antigen RoTAT1.2 with 205 bp of weight is attached to pET22b (+) vector and expressed in E.coliBL21 system (DE3). RoTAT1.2 recombinant antigens with weight approximately 12 kDa recognized by T.evansi specific antibodies. The suitable conditions for recombinant antigen is RoTAT1.2: IPTG induction concentration: 0.1 mM, induction time: 5 hours, sensor temperature around 30 ° C. The results of this study are the basis and materials to produce RoTAT1.2 recombinant antigen, serve for manufacturing of bio Trypanosomiasis in Vietnam’s cattle. |
